https://www.newscientist.com/article/2095769-gut-bacteria-spotted-eating-brain-chemicals-for-the-first-time/ 제가 장내세균에 관심이 많아요. 똥캡슐은 언젠가 빛을 보고 말리니. 2011년에 이미 장내세균과 뇌의 연관성, 그로 인한 기분변화에 대한 논문이 발표되었었죠. 이번에 자세히 보니 뱃 속의 어떤 세균이 우리 뇌속의 기분을 좌우하는 중요한 화학물질을 먹고 살더란 얘기. 이 세균의 이름은 KLE1738이에요. 문득 제가 참 좋아했던, 전형적이고 유치했지만 재밌었던 헐리우드 액션영화 < 히트맨 >이 생각나는군요. 거기 나오는 전문암살요원 주인공 이름이 42번이었어요. No. 42. 남주를 맡은 배우가 Timothy Olyphant예요. 여주는 유명한 듯한 섹시러시아배우 올가였어요. 몸매가 정말 이쁘죠. 영화장면에 이런 게 나와요. 올가가 42번에게 납치되어 호텔이며 차며 강제인 듯 아닌 듯 끌려댕겨요. 어느날 호텔에서 옷을 훌러덩 벗고 42번의 넥타이를 확 끌어당기며 유혹을 합니다. 42번은 표정 하나 안 변하고 무뚝뚝한 얼굴로 여자를 휙 밀어버리는데 얼마나 가차없이 훌떡 밀어버리던지 그게 참 재밌고 멋있었어요 ㅋㅋ. 여튼 다시 장내세균으로 돌아와서, KLE1738에게 GABA라는 분자물질을 제공해 주었더니 이걸 먹고 살더란 얘기죠. GABA는 신경세포로부터 전해져 오는 시그널을 차단해주는 역할을 하거든요. 그로 인해 우리의 뇌는 안정하게 돼요. GABA가 많이 없으면 사람은 기분이 우울하게 됩니다. 연구팀은 GABA를 먹는 다른 박테리아나 심지어는 GABA를 생산하는 박테리아가 있나 살펴볼 거예요. 그리고 그런 장내세균이 우리의 뇌에 끼치는 영향을 알아볼 생각이죠. 우울증을 치료할 획기적인 약은 개발할지 못한다 하더라도 인간의 삶을 조금이나 개선해줄 묘책을 발견할 수 있다면 좋겠어요. 알파 아미노산은 쭉 이어져서 단백질을 구성하는데 감마 아미노산은 딱 세 칸 떨어져 있다고 생체 내에서 완전 다른 역할을 합니다. 뇌세포와 각종 신경 세포에서 신경 전달 물질(neurotransmitter)로 작용하는 거죠. 단순한 분자인 만큼 뭔가 큰 비밀을 가지고 있을 것 같은 느낌이 있죠. 이 사실이 밝혀지고 난 뒤 생리학자들과 약리학자들, 거대 제약회사들이 GABA에 큰 관심을 가지고 연구를 많이 해서 엄청나게 많은 지식을 얻었어요. 그래서 괜히 GABA를 세균한테 먹이로 줘보기도 하는 거지요. 보통 우리 주변에 흔한 세포들은 아미노산, 포도당, 지방산을 연료로 살아가는데요. 가끔 다른 환경에 살거나 특이한 조건에서는 다른 분자를 연료로 쓰는 경우가 있어요. 이 세균도 GABA를 줬더니 연료로 쓸 수 있다는 것이죠.
GABA는 생체 내에서 합성하기도 하지만 연료로 쓰는 것이 아닌 만큼 많이 필요로 하진 않아요. 대신 음식물에서도 섭취할 수 있는 것이죠. 음식물이 단백질, 탄수화물, 지방으로만 되어 있는 건 아니겠죠. 동식물들을 먹으니 GABA도 당연히 먹게 되죠. 어떤 세균이 GABA를 빼앗아 먹게 되면 GABA가 부족해지고 이것이 뇌에 영향을 줄 수도 있지 않을까 하는 약간 소설 같은 논문이지요.
그렇다면 KLE는 무엇인가, 아마 Klepsiella 같은데요. 대표적인 혐기성 세균이자 장내 세균입니다. 혐기성이란 산소로 호흡하지 않기 때문에 산소를 싫어한다는 뜻인데요. 장내라니 공기가 안 통할 것 같지요. 그래서 장내 세균은 대부분 혐기성 세균이고 장 바깥 세상과 다른 자기들만의 생태계를 구성합니다. 이것이 장내 세균총(flora)이고 눈부심님이 관심 있어하는 애들이죠. 얘네 중에 어떤 애들은 GABA를 밥 대신에 먹는다는 겁니다.
그런데 이 장내 세균총이란 의학적으로 상당히 중요하면서도 위험한 애들입니다. 항생제를 너무 많이 써서 장내 세균총이 약해져 버리면 다른 강한 세균이 얘네가 차지하고 있던 자리를 대신 차지해 버리면서 심각한 장염을 일으킵니다. Clostridium difficile이란 균인데요. 이걸 치료하려고 다른 사람의 대변을 이식해서 장내 세균총을 회복시키고 C. difficile을 밀어내려는 것이죠. 그런데 사실 이 C. difficile의 위험보다 더 무서운 것이 있습니다. 장내 세균총 그 자체이죠. 맹장염이 심해져서 맹장이 터지게 되면 장에 있던 내용물이 주변 배 속 공간으로 다 튀게 되는데 장내 세균총에 의해 감염이 빠르게 진행되면서 위험해지는 것입니다.
뿐만 아니라 항생제를 지속적으로 먹는 사람의 장내 세균총은 계속 살아남으면서 항생제에 내성을 갖게 됩니다. 이렇게 자란 균은 죽이기가 어렵죠. 한국에서 Klepsiella는 아주 높은 빈도로 많은 항생제에 내성을 가지고 있습니다. 이게 더 자라면 슈퍼 박테리아가 되는 것인데 이런 균은 침도 제대로 못 삼키고 기침을 계속 하면서 기도에 관을 넣고 호흡기를 달고 있는 환자들이 많은 중환자실에서 아주 아주 위험하겠죠. 건강한 사람에게는 건드리지 않고 사이 좋게 지내야 하는 게 장내 세균총이지만 함부로 건드리면 위험해요. 장내 세균총 너무 좋아하지 마세요~.
